LightCycler® Uracil-DNA Glycosylase
LightCycler ® 尿嘧啶-DNA糖基化酶水解單鏈和雙鏈DNA中U-DNA位點(diǎn)的尿嘧啶-糖苷鍵,切除尿嘧啶并在DNA中產(chǎn)生對堿和熱敏感的脫堿基位點(diǎn)。
該酶對單鏈 DNA 的活性高于對雙鏈 DNA 的活性
該酶對 RNA 和天然無尿嘧啶 DNA 無活性。

The enzyme is supplied in storage buffer at 2 U/µl.

Application應(yīng)用:
LightCycler® Uracil-DNA Glycosylase (LightCycler® UNG) is ideally suited for preventing carryover contamination between PCRs. This technique involves incorporating deoxyuridine triphosphate (dUTP) into all amplification reactions, and pretreating all successive LightCycler® FastStart Enzyme-based PCR mixtures with LightCycler® UNG. Prior to the thermal-cycling procedure, LightCycler® UNG is added to the LightCycler® amplification reaction mixture, then incubated for 10 minutes at +40°C. During the initial activation of FastStart DNA Polymerase (+95°C for 10 minutes), the LightCycler® UNG becomes inactive.
